【 Test principle 】

Hematoxylin-eosin staining solution (H-E strain) is mainly used to display the normal components of various tissues and the general morphological structure of lesions for comprehensive observation. Hematoxylin-eosin staining solution is the most essential staining method in biology, histology, pathology and cytology. It is widely used in pathological diagnosis, teaching and scientific research, and has important value. The nucleus of the cell is composed of acidic substances, which have a strong affinity with the basic dye (hematoxylin), while the cytoplasm, which contains alkaline substances and an acidic dye (eosin), has a strong affinity. Therefore, after the cells or tissue sections are stained by hematoxylin and eosin staining solution, the nucleus is stained by hematoxylin into a bright blue-purple color, the cytoplasm, muscle fibers, collagen fibers, etc., are red in varying degrees, and the red blood cells are orange-red.

【 Note 】

1. It is normal to produce an oxide film on the surface of hematoxylin dye solution and a little aluminum sulfate crystal precipitation at the bottom. The oxide film should be removed before use.

The diluted lithium carbonate solution is then used for bluing.

2, when the temperature is low, the hematoxylin dye is not easy to color, and the dyeing time can be appropriately extended.
